A method for the determination of C-terminal amino acids and C-terminal amino acid sequences in nanomolar amounts of proteins is described, based on carboxypeptidase A digestion of the protein, followed by removal of the partially digested protein and quantitative gas-liquid chromatographic determination of the amino acids released after known time intervals. Sequences deduced from the kinetics of release of specific amino acids are compared with the known C-terminal sequences of well-characterized proteins.  相似文献

A rapid and efficient total synthesis is reported for the cyclic lipodepsipeptide pseudodesmin A. This member of the Pseudomonas viscosin group is active against Gram‐positive bacteria and features self‐assembling properties. A conserved serine residue within the lactone macrocycle is exploited for initial immobilization on 2‐chlorotrityl chloride resin through ether formation with the side‐chain alcohol. Subsequent elongation proceeds through Fmoc solid‐phase peptide synthesis, including automated incorporation of the enantioselectively synthesized (R)‐3‐hydroxydecanoic acid lipid tail. Following esterification to generate the incipient lactone bond, the macrocycle is formed by on‐resin head‐to‐tail macrolactamization and cleaved from the resin to give the desired compound in good purity. The short and efficient synthesis route allows rapid generation of analogues by facile variation of both the peptide and lipid moieties with good control of epimerization while maximizing automation. Synthesis of the pseudodesmin A enantiomer yields identical self‐assembly and biological activity to that observed for the natural compound, showing that activity is not mediated by chiral interactions. A D ‐Asn8 analogue developed en route retains self‐assembly, but loses activity. The synthesis strategy should be generally applicable for the rapid generation of analogues from various cyclic lipodepsipeptide groups, allowing an investigation of their self‐assembling properties and structure–activity relationships.  相似文献

CZE is an appropriate technique for separating charged species, but lacks selectivity for neutral compounds. Alternative approaches such as microemulsion electrokinetic chromatography (MEEKC) have been developed to broaden its range of applications. Hyphenation of MEEKC with MS is an attractive perspective since it can enhance sensitivity and selectivity. The on-line coupling of MEEKC with MS, however, is not straightforward due to the low compatibility of non-volatile surfactant additives (e.g. SDS) and the commonly used API source, namely ESI. In order to hyphenate MEEKC with MS detection, the atmospheric pressure photoionization (APPI) source was investigated. Possibilities offered by the coupling of MEEKC with APPI-MS were highlighted for the complex separation of ionized and neutral compounds in both the positive and negative modes. MEEKC-APPI-MS performance, in terms of selectivity, efficiency and sensitivity was compared to CZE-ESI-MS and MEEKC-ESI-MS for the screening of doping substances (beta-blockers, central stimulants, diuretics, etc). Relevant selectivity and detectability, particularly for neutral, structurally related and isobaric compounds was demonstrated with the MEEKC-APPI-MS approach opening new avenues for CE-MS, in addition to the well-established CZE-ESI-MS technique.  相似文献

The sorption behavior of three benzyl alcohol derivatives with different hydrophobicities into sonicated and extruded DODAC dispersions has been studied using NMR spectroscopy and NMR diffusometry. We show that there is an increased sorption into a sonicated dispersion below the phase-transition temperature (T(m)) as compared to an extruded dispersion. This may be explained by the incomplete lipid chain freezing of charged lipids as a result of the sonication process. Around T(m), a sorption maximum is found that is attributed to the high bilayer disorder under this condition. In addition, a sorption increase and a fluidizing effect at increasing benzyl alcohol derivative concentrations are observed that provide additional evidence for the relevance of the bilayer fluidity on the sorption of hydrophobic components.  相似文献

In view of efficient and nontoxic delivery of genes to cells, complexes made of phospholipids (noncationic) and DNA are assembled through the mediation of multivalent cations. The association of lipids with DNA is explained through the charge reversal of lipid headgroups by specific adsorption of cations. The ion binding is quantified by the Gouy-Chapman-Stern theory which provides a good estimate for the minimal concentration of cations required to produce complexes. Coarse-grained Monte Carlo calculations support X-ray diffraction experiments in the sense that lipids form inverted micelles around hexagonally arranged DNA rods, with cations in between to maintain the cohesion. The complexes are more cohesive in terms of total free energy as the cation valence increases. The presented methodology may help develop predictive models for biomolecular self-assembled systems.  相似文献
